dc.description.abstractO nitrogênio é o nutriente requerido em maior quantidade pelas plantas e a utilização de fertilizantes nitrogenados agrava os problemas ambientais, além de gerar alto custo na produção. A exploração da simbiose entre leguminosas e bactérias fixadoras de nitrogênio atmosférico pode aumentar a produtividade das culturas e reduzir os custos de produção. Os objetivos desse trabalho foram avaliar estirpes de rizóbios isoladas de feijão-caupi e soja quanto a capacidade de nodulação cruzada entre estas espécies e eficiência na fixação biológica do nitrogênio e caracterizá-las por meio do sequenciamento dos genes 16S rRNA, recA, gyrB e nodC. Inicialmente foi realizado um experimento de autenticação da capacidade de nodulação de 50 estirpes de rizóbio isoladas de feijão-caupi e soja do campo experimental da Embrapa Agrobiologia, no município de Seropédica e inoculadas de forma cruzada nestes hospedeiros. Deste experimento foram selecionadas as estirpes BR 10737 e BR 13971 para um segundo experimento da avaliação da eficiência simbiótica em feijão-caupi, e as estirpes BR 13895, BR 13971 e BR 13986, para da soja. Estas bactérias foram testadas em um experimento de eficiência simbiótica comparando-as com as estirpes BR 3262 e BR 29, recomendadas pelo MAPA para inoculação de caupi e soja, respectivamente. Os números de nódulos (NN), massa seca de nódulos (MSN) e massa seca da parte aérea (MSPA) foram avaliados. Foi observado que 31 estirpes nodularam o feijão-caupi e 11 a soja, demonstrando que esta leguminosa apresenta maior especificidade hospedeira. As estirpes BR 10737 e BR 13971, apesar de contribuírem para o crescimento das plantas, não apresentaram médias significativas de NN, MSN e MSPA em comparação com a inoculação padrão. As estirpes BR 13895 e BR 13971 também apresentaram médias significativamente inferiores para a maioria das variáveis na inoculação da soja. Somente a BR 13986 mostrou potencial para ser utilizada como inoculante para a cultura da soja, visto que suas médias foram semelhantes às da estirpe recomendada BR 29. Em seguida, foi realizada a amplificação e sequenciamento dos genes 16S rRNA, recA, gyrB e nodC, e a construção das árvores filogenéticas. Com base nos resultados concluiu-se que as 50 estirpes pertencem gênero Bradyrhizobium e há uma alta diversidade entre as estirpes, com a indicação de novas espécies. Apesar das estirpes estarem estruturalmente associadas e filogenéticamente próximas pelo gene 16S rRNA, apresentam genes nodC bastante distintos. Foi concluído que há potencialmente cinco novas espécies, sendo quatro isoladas de feijão- caupi e uma da soja e com estirpes com potencial para testes como inoculantes, sobretudo para a soja.pt_BR

dc.description.abstractOtherNitrogen is the nutrient required in greater amount by plants and the use of nitrogen fertilizers aggravates environmental problems, in addition to generating high production costs. Exploring the symbiosis between legumes and atmospheric nitrogen-fixing bacteria can increase crop yield and reduce production costs. The objectives of this work were to evaluate rhizobia strains isolated from cowpea and soybean in terms of cross-nodulation capacity between these species and efficiency in biological nitrogen fixation, and to characterize them through the sequencing of the 16S rRNA, recA, gyrB and nodC genes. Initially, an experiment was carried out to authenticate the nodulation capacity of rhizobia 50 strains isolated from cowpea and soybeans in the experimental field of Embrapa Agrobiologia, in the municipality of Seropédica, and cross-inoculated in these hosts. From this experiment, strains BR 10737 and BR 13971 were selected for a second experiment to evaluate the symbiotic efficiency in cowpea, and strains BR 13895, BR 13971 and BR 13986, for soybean. These bacteria were tested in a symbiotic efficiency experiment comparing them with strains BR 3262 and BR 29, recommended by MAPA for cowpea and soybean inoculation, respectively. The number of nodules, dry mass of nodules and dry mass of shoots were evaluated. It was observed that 31 strains nodulated cowpea and 11 the soybean, demonstrating that this legume has greater host specificity. Strains BR 10737 and BR 13971, despite contributing to plant growth, did not show significant means of NN, MSN and MSPA compared to the standard inoculation. Strains BR 13895 and BR 13971 also showed significantly lower means for most variables in soybean inoculation. Only BR 13986 showed potential to be used as an inoculant for the soybean crop, since its averages were similar to those of the recommended strain BR 29. Then, the amplification and sequencing of the 16S rRNA, recA, gyrB and nodC genes was carried out, and the construction of phylogenetic trees. Based on the results, it was concluded that the 50 strains belong to the genus Bradyrhizobium and there is a high diversity among the strains, with the indication of new species. Despite the strains being structurally associated and phylogenetically closed by 16S rRNA gene, they present quite distinct nodC genes. It was concluded that there are potentially five new species, four isolated from cowpea and one from soybean and with strains with potential for testing as inoculants, especially for soybean.en
